Do I have to accept the highway agency's first offer?

No. The offer from TxDOT, FDOT, Caltrans, NCDOT, or any other highway agency is built from their own appraisal and is an opening position, not a final number. You have the right to an independent review and to negotiate before you sign anything.

What if only part of my property is being taken for the highway?

In a partial taking, you're owed for the strip of land taken plus the damage done to the remainder — including lost access, reduced frontage, and diminished use of the land you keep. That remainder damage is often far larger than the value of the strip itself and is commonly underestimated in the initial offer.

Do the same compensation rules apply to toll roads and county or city road projects, not just state DOTs?

Yes. Toll authorities, county governments, and municipalities follow the same just-compensation rules as state DOTs when they condemn property for a road project, so the same right to an independent review and negotiation applies.
